Effects of substance P on neuronal firing of pallidal neurons in parkinsonian rats.

چکیده
Substance P is an important neurotransmitter or neuromodulator in central nervous system. Morphological studies have revealed the existence of substance P and its high affinity receptor, neurokinin-1 receptor, in globus pallidus. The expression of neurokinin-1 receptor in external globus pallidus has been reported to be decreased or unchanged in parkinsonian patients. To further investigate the effects of pallidal neurokinin-1 receptor in Parkinson's disease, an in vivo extracellular recording in 6-hydroxydopamine parkinsonian rats was performed. Micro-pressure ejection of selective neurokinin-1 receptor agonist, [Sar9,Met(O2)11] substance P (0.1mM), increased the spontaneous firing rate of pallidal neurons by 9.1% on the lesioned side, which was significantly weaker than that on the unlesioned side (20.7%), and that in normal rats (30.0%). The selective neurokinin-1 receptor antagonist, SR140333B, prevented the excitatory effects induced by [Sar9,Met(O2)11] substance P. Based on the action of substance P in globus pallidus of parkinsonian rats we hypothesize that the activity of neurokinin-1 receptors in globus pallidus may be decreased under parkinsonian state. This finding may provide a rationale for further investigations into the potential of pallidal substance P system in the treatment of Parkinson's disease.

The effect of intrastriatal injection of estrogen on pallidal field potential and rigidity in Parkinsonian -ovariectomized rats
The major pathological feature of Parkinson’s disease (PD) is a progressive loss of dopamine-producing neurons of the substantia nigra pars compacta (SNc), resulting in a reduction of dopamine (DA) content in the target field of these neurons, the striatum (STR). The present evidences suggest that female sex hormones may influence the onset and severity of PD symptoms. PD is more prevalent in m...

متن کاملEffects of Pharmacological Block of GABAA Receptors on Pallidal Neurons in Normal and Parkinsonian State
The globus pallidus plays a central integrative role in the basal ganglia circuitry. Morphological studies have revealed a high level of GABA and GABA(A) receptors in the globus pallidus. To further investigate the effects of endogenous GABA(A) neurotransmission in the globus pallidus of normal and parkinsonian rats, in vivo extracellular recording and behavioral tests were performed in the pre...
متن کاملEffects of resveratrol on intrinsic neuronal properties of CA1 pyramidal neurons in rat hippocampal slices
Introduction: Resveratrol (3,5,4-trihydroxystilbene) a non-flavonoid polyphenol found in some plants like grapes, peanuts and pomegranates, possesses a wide range of biological effects. Evidence indicates that resveratrol has beneficial effects on nervous system to induce neuroprotection. However, the cellular mechanisms of the effects are not fully determined. In the present study, the cellula...
متن کاملEffect of aqueous extract of Drosera Spatulata on firing rate of paragigantocellularis nucleus neurons after pain induction by formalin in rats
Introduction: Previously, we demonstrated that i.p. injection of aqueous extract prepared from aerial parts of Drosera Spatulata (Droseraceae) can induced remarkable analgesia in both phases of formalin test in rats. Because, analgesia induced in acute phase of formalin test mainly mediated by activation of central analgesic mechanisms and also paragigantocellularis (PGi) nucleus is part of bra...
